FACILITIES

The BCRI has a state-of-the-art Coulter Elite ESP three laser flow cytometer (left). The Elite flow cytometer/sorter provides three color laser excitation and 7 parameter list-mode analysis for each cell at rates of 1 x 103 cells per second. The systems also allow for sorting and contains an Autoclone setup which allows sorting of individual cells into individual wells of a 96 well plate. 

  

 

The FACScan flow cytometer is ideal for obtaining three color surface immunofluorescence.

In addition there is a  Laser Scanning Cytometer (CompuCyte Corp., Cambridge, MA)  (left) that allows for laser illumination of cells stained on slides. This system is capable of measuring 7-11 parameters per cell at rates of several hundred cells per second and contains several unique features: 1) Merge allows the same cell to be measured several times (with different dyes or live and then fixed) with all data stored in list mode for multiparameter analysis. (This system records the position of each cell so that multiple rounds of analysis are possible). Merge also allows the same cell to be measured several times in quick succession for detecting enzyme kinetics, etc; 2) Maximal Pixel measurements allows for not only determination of the amount of fluorescence associated with each cell but also provides information on the change in distribution of material within the cell - maximal pixel refers to the intensity of localized fluorescence allowing for observations of accumulation of material in specific intracellular organelles; and Separate thresholding for nucleus and cytoplasm allows for the analysis of traffic of molecules into and/or out of the nucleus since both the fluorescence associated with both the nucleus and cytoplasm can be analyzed separately. In addition, the system is equipped with a high resolution color CCD camera which is used to image cells selected based on any combination of fluorescence measurements and provides a gallery of such cells for any subpopulation desired.The BCRI also has two separate tissue culture facilities containing laminar flow hoods, CO2 incubators, refrigerators/freezers, liquid nitrogen for storage of cell lines, etc. A separate molecular biology facility is also present which contains 3 PCR systems, several gel electrophoresis systems (Wester, Northern, Southern and Pulse field), Speedvac concentrators (2), Nikon Microphot FXA microscope (with two 35 mm and one 4 x 5 format cameras and both fluorescence and interference optics), electroporation apparatus, desk-top publishing system, and refrigerators/freezers, balances, centrifuges (refrigerated and room temperature) normally found in laboratories. The BCRI provides additional services and instrumentation. Thus, as noted above, there is a dark room for processing of film containing a Kodak M35A X-OMAT film processor.
